Nootropic drugs, such as aniracetam, etiracetam, nefiracetam, oxiracetam,  piracetam, pramiracetam,  and rolziracetam are used in the treatment of dementia and cognitive enhancement. These nootropic agents were discovered in the 1960s.

The effectiveness of Oxiracetam (1600 mg/day) versus placebo was assessed in a group of 96 out-patients suffering from cognitive disorders secondary to primary degenerative dementia. The study lasted twelve months and was performed in two stage, a) double-blind (26 weeks) and b) open study (26 weeks). The patients treated with Oxiracetam showed a statistically significant improvement of simple reaction time and cognitive function detected by the Attention matrix. In the placebo group after twelve months a significant worsening of cognitive and global function was observed in comparison with baseline scores. The patients themselves appeared in favor of Oxiracetam. The drug tolerability proved to be very good for the whole duration of the treatment. The authors believe that Oxiracetam favorably acts on the symptoms of senile cerebral deterioration and can improve the capability of information processing, as suggested by the better performances obtained at the reaction time test and at the Attentional Matrix test.

Animal studies with Oxiracetam
Oxiracetam prevented the scopolamine but not the diazepam induced memory deficits in mice.

In mice, the elevated plus-maze paradigm was used to investigate the effect of scopolamine hydrobromide and diazepam and their interaction with oxiracetam on the retrieval of spatial memory trace. Experiment 1: Scopolamine  and diazepam given 30 min before the retention session significantly prolonged the transfer latency as compared with the saline treated mice and those given the lowest dose of scopolamine and diazepam. Experiment 2: Oxiracetam administered immediately after the acquisition session prevented the scopolamine induced prolongation of the transfer latency. Thus, oxiracetam forestalled the impairment of retrieval of memory trace: the animals were able to remember the spatial configuration of the plus-maze. On the contrary, oxiracetam was not effective in the diazepam treated mice. We suggest that beneficial effect of oxiracetam might be confounded or blocked by the anxiolytic effect of diazepam.

Q. What are your thoughts on the efficacy of oxiracetam? I've been taking an 800 mg capsule 3-4 times a week for four years and definitely notice an improvement in mental functions such as quicker thought processes and improved memory.
   A. I prefer to use natural supplements since they provide good benefits. I personally have not tried oxiracetam.
